Genetic information | Mice express Acot7 cDNA under the control of a tetracycline-inducible bidirectional promoter. Firefly luciferase is expressed simultaneously under the control of the same promoter. Insertion was via pronuclear injection into C57BL/6J pronuclei; integration was aleatory - we do not know the location in the genome. Phenotypic information | Homozygous:We have not studied homozygous mice. No phenotype should be observed in the absence of further breeding with animals expressing rtTA and in the presence of doxycycline.Heterozygous:When bred with animals expressing rtTA specifically in pancreatic beta cells, and upon doxycycline administration, the mice develop mild glucose intolerance, impaired insulin secretion in response to glucose and the islets present reduced levels of fatty acid-CoA. |
Breeding history | The founder animal was first bred with C57BL/6 wild-type, and the F1 again with C57BL/6 wild-type to confirm only one insertion by qPCR. F2 was bred with littermate controls (not containing the transgene) or C57BL/6 wild-type for at least 4 generations. |
