Phenotypic information | Homozygous:Mice hemizygous for the Nr5a1::hM4D-2A-TomatoF BAC transgene are viable and fertile, with expression of the pharmacogenetic inhibitory receptor hM4D (Armbruster et al. 2007) and farnesylated Tomato in the dorsomedial portion of the ventromedial hypothalamic nucleus (VMHdm). The expression of hM4D and TomatoF is restricted to Nr5a1 expressing cells. The hM4D is a modified version of the human muscarinic receptor M4 with high affinity for the otherwise biologically inert drug Clozapine-N-Oxide (CNO), and virtually no affinity to its endogenous ligand acetylcholine. Activation of the hM4D by CNO in neurons induces a decrease in membrane potential and neuronal firing and was used to selectively and rapidly inhibit neurons in the VMHdm in behaving animals.
