Genetic informationThe human TNF/beta-globin (TNF-globin) recombinant gene construct was prepared in the pBluescript vector as follows: The 2.8 kb EcoRI fragment which contains the entire coding region of the human TNF gene was ligated to the 0.77 kb EcoRI-Sall fragment which contains the 3'-untranslated region and polyadenylation site of the human beta-globin gene (Kollias et al., 1987) and excised for microinjection as a BamHI-Sall fragment.
Phenotypic informationTransgenic mice expressing human tumour necrosis factor: a predictive genetic model of arthritis. In contrast to the other line expressing the same transgene (Tg(TNF)197Gkl), the onset of arthritis occurs later on in adult life.
