Genetic information | Bacterial artificial chromosome (BAC) library was used to obtain a 130 kb BAC containing the entire mouse Trp53 (transformation related protein 53) gene. This BAC also contains other active genes such as Efnb3 (Ephrin B3). The BAC was microinjected into fertilized (C57BL/6 x CBA)F1 oocytes. Super p53 founder, carrying 1 copy of the BAC, was established and then subsequently maintained by breeding transgenic mice with C57BL/6 inbred mice for at least 10 generations. |
Phenotypic information | Mice hemizygous for the BAC Super p53 transgene are viable and fertile. These mice express one extra copy of the wild-type mouse Trp53 (transformation related protein 53) gene directed by its endogenous promoter/enhancer regions on the BAC transgene. Trp53 encodes a tumor suppressor protein, which also functions as a transcription factor. It can trigger proliferation arrest and cell death in response to stresses such as DNA damage and oncogenic stress. Super p53 mice show an enhanced response to DNA damage and they are protected from cancer. They also age normally in comparison to other p53 over expressing mouse lines. |
Breeding history | Founder was crossed with C57BL/6J animals. The line was backcrossed into C57BL/6J for more than 10 generations. The line is currently maintained by homozygous crosses. |
