Genetic information | Regulated expression of a truncated, constitutively active form of calcineurin (CnA or Ppp3ca) in astrocytes was achieved with the Tet-off system by generating two transgenic mouse lines. A first mouse line expressed tet-transactivator (tTA) under the astrocyte-specific GFAP promoter. A second transgenic line expressed CnA under the control of the TetO promoter. A cDNA encoding a truncated form of the murine calcineurin catalytic subunit A, CnA (OKeefe et al., 1992), was placed downstream from the TetO promoter (from pTRE vector; Clontech). The pGFAPtTA and TetOCnA DNA fragments were purified and independently injected into fertilized eggs of C57BL/6 x SJL mice. Founder mice were analyzed by Southern blotting and PCR (see Fernandez et al., 2001), using specific primers and backcrossed to C57BL/6 mice to generate the lines pGFAPtTA (GFAP) and TetOCnA (CnA). The astrocyte inducible calcineurin (AIC) mice were generated by crossing the GFAP7 (F3) with the CnA5 (F3) mice. |
