Genetic information | Transgenic mice for a U6-shRNA gene. 13 copies of transgene. The U6-shRNA construct encompasses the U6 mouse promoter that drives the transcription of a shRNA expressing transcription unit. The shRNA sequence targets the 3'-part of the intermediate early gene (IE gene) of the Pseudorabies virus (PRV), with the following sequence (5'-CTCCCATCGGGTGAGAAAA-3'). PRV currently infects farm animals (especially pigs) and mice. The U6-shRNA gene is inserted in an empty transgene expression vector encompassing the 5'HS4 insulator from the chicken beta globin gene locus, the human eF1alpha gene promoter (-1430, +2), the rabbit (rb) beta-globin second intron and several enhancers (E1, E2 and E3). The empty expression vector acts as a cargo for the U6-shRNA gene to insure the reliable expression. |
