Genetic informationThe complete cDNA sequence for human ELAVL1 (HuR) was obtained from the IMAGE clone number 2901220 (GenBank accession number BC003376) and was subcloned in-frame to an HA-epitope tag for the expression of an N-terminally tagged form. Subsequently, the HA-HuR cDNA was used for the generation of a transgenic construct that contained, in a 5' to 3' orientation, seven copies of the 42 bp tet operator sequence upstream of the minimal CMV promoter (Clontech); a globin intronic sequence for optimal transgene expression; the HA-HuR cDNA; and a bovine growth hormone polyadenylation signal. For the production of transgenic mice, fertilized (CBA x C57BL/6)F2 hybrid zygotes were coinjected with the transgenic devices using standard procedures.
