Genetic informationThe genomic locus containing the genes for Tnf and lymphotoxin a, was isolated from a 129Sv mouse genomic library in lamdaFIXTMII. The replacement-type targeting vector was made by replacing the 724 bp BglII-SmaI fragment containing the second and third exons of Tnf with a mutated 511 bp BglII-SmaI fragment carrying a deletion of the first twelve amino acids of soluble Tnf (Tnf delta1-12). A pL2neo gene flanked by loxP sites was inserted into the first intron of the Tnf gene at the BglII site, and a PGK tk gene was inserted at the 5' end of the targeting vector for negative selection. The targeting vector contained 4.9 kb homologous DNA upstream of the first loxP site and 0.7 kb homologous DNA downstream of the Tnf delta1-12 deletion. The targeting vector was electroporated into CCE ES cells.
Phenotypic informationMutant mice produce physiologically regulated levels of transmembrane TNF (tmTNF), but cannot substitute for the hepatotoxic activities of wild-type TNF following LPS/D-galactosamine challenge in vivo and are not sufficient to support secondary lymphoid organ structure and function.
