Genetic information | Mice express the herpes simplex virus thymidine kinase (HSVTK) under the Itgam promoter. This allows for the conditional depletion of macrophages/microglia by administration of the thymidine analogue ganciclovir (GCV). |
Phenotypic information | In vivo administration of GCV to CD11b-HSTK mice leads to aplastic anemia with 5-10 days by depletion of hematopoietic precursor cells. For short term experiments, mice can be reconstituted with wild-type bone marrow and treated systemically with GCV to deplete radioresistant microglia. For ex vivo cultures, no adverse effect on other cell types when depleting microglia. |
Breeding history | Mice have been maintained on a C57BL/6JHsd background for at least 10 generations. Males are infertile due to the expression of the long form of the HSVTK enzyme. Females are fertile, but tend to give small litters of 5-6 pups. |
