Genetic informationThe cre recombinase gene was cloned under control of the human C-reactive protein (hCRP) promoter. The hCRP-promoter is inducible by lipopolysaccharide (LPS) or interleukin 6. Upon induction the cre gene is strongly expressed in hepatocytes, exclusively. The amount of cre-mediated deletion can be controlled by the amount of LPS injected. The percentage of deletion is in the range of 13 to 45%. Exon V of Mgmt was flanked by two loxP-sites. This exon can be spliced out by cre recombinase thereby inactivating the DNA repair activity of the MGMT protein.
Phenotypic informationNo major defects observed. The Tg(CRP-cre) transgene allows the liver-specific, inducible expression of cre recombinase.
