Genetic information | The pHGStopPrP-Fc construct was used for nuclear injection, with a loxP-flanked transcriptional stop cassette (pBS302, Invitrogen) inserted upstream of the PrP-Fc ORF. The generated Tg588 line can be bred with a cre recombinase-expressing line to enable ubiquitous or tissue-specific expression of PrP-Fc. |

Breeding history | A founder of the Tg588 line was bred with a C57BL/6 mouse. Breeding of transgene-hemizygous animals (with wild-type Prnp +/+ genotype) with C57BL/6 mice continued for more than 5 generations. The Tg588 transgene-positive mice are viable and without any defect also on the Prnp -/- (129/Sv x C57BL/6) background. |
