Genetic informationA loxP site was inserted 3' of exon v6 and a neomycin resistance cassette, with a 5' loxP site, was inserted 5' of exon v6 of the Cd44 gene. During insertion, 1.6 kb of sequence was lost in the exon v7 region leading to deficient expression of this variant in homozygous mutant mice (Cd44v7 -/-). Insertion of a neomycin resistance cassette into the unique exon of the Rag2 gene by homologous recombination leading to the deletion of 286 amino acids.
Phenotypic informationRag2-/-: No mature T and B cells. Cd44v7-/-: Strongly reduced symptoms in autoimmune diseases.
