Genetic information | The wild-type Fas ligand (Fasl) allele was replaced with a deletion mutant which lacks the intracellular domain (i.e. aa 2-73). The targeting construct contained a tk-neo fusion cassette flanked by loxP sites for positive selection. This tk-neo cassette has been removed after breeding with CMV-cre recombinase deleter mice. |
Phenotypic information | Homozygous mutants develop a lymphoaccumulation of T-cells in the periphery, similar to Fas ligand-deficient gld/gld mice (of a strain with a Fas ligand spontaneous mutant allele). In addition, mice probably develop autoimmune defects, again similar to gld/gld mice. Therefore, homozygous mice die within 4-6 months of age in the C57BL/6 background. |
