Genetic information | In order to obtain tetracycline-controlled expression of the human cystic fibrosis transmembrane conductance regulator (CFTR), the CFTR cDNA was cloned downstream of the conditional promoter PhCMV*-1 in the plasmid pUHD.10-3. Three transgenic lines were obtained with pUHD.CFTR: TC3, TC7 and TC35. These lines were backcrossed with CBA/Ca mice for several generations, as this strain has been observed to improve expression from transgenics using the tTA inducible system. They were then crossed with JY13 mice which carry a transgene consisting of the tTA open reading frame driven by the promoter of PMP22 (peripheral myelin protein 22), which is highly expressed in the Schwann cells of the peripheral myelin system. They were also crossed with JU2 mice which carry the lacZ gene under the control of the conditional promoter PhCMV*-1. This allows assessment of tTA induction of gene expression. |
