Genetic informationGeneration of the rhombomere (r)4-cre transgenic line: A 2,7 kb XhoI-HindIII construct containing the cre recombinase gene and the metallothionein-1 polyadenylation signal obtained from pBS185 (GibcoBRL, Cat# 10347-011) was cloned into EcoRV -SacII of pWexP3β (Danielian and McMahon 1996; gift from Paul S. Danielian), which includes a Wnt1 basic promoter. The 741 bp ApaI-SpeI fragment containing the Hoxb1 r3/r5 repressor and the 1043 bp SpeI-SmaI fragment containing the Hoxb1 5' r4 enhancer were cloned inverted (Studer et al. 1994) and inserted into the BglII of the above plasmid. This combination leads to restricted expression of cre recombinase in rhombomere 4 of developing embryo hindbrain. The targeting construct was digested with NotI and injected into pronuclei of CD1 oocytes.
