Genetic informationThe endogenous Trp53 allele carries a point mutation, TCC to GCC in exon 5, resulting in Ser to Ala substitution at codon 180.
Phenotypic informationHomozygous:
Homozygous S180A mutant mice are tumor resistant but have slightly shortened lifespan. For example, S180A knock-in mice showed markedly reduced spontaneous tumorigenesis and increased resistance to Myc-driven lymphoma and Eml4-Alk-driven lung cancer. Median lifespan was significantly reduced from 709 days in wild type to 568 days in S180A littermates. Lifespan was reduced by a loss of general fitness and increased susceptibility to age-related diseases.

Heterozygous:
Heterozygous mice have normal phenotype.
Breeding historyS180A mutant mice were originally maintained in a 129 background (and also archived in this background, see EM:13426). They were subsequently backcrossed to C57BL/6J background for 10 generations.

Requesting frozen sperm or embryos is generally advisable wherever possible, in order to minimise the shipment of live mice.
