Phenotypic information | Homozygous:Two Scapers homozygous mutant males were fertile in the C57BL/6J genetic background. This contrasts with the sterile phenotype observed on the original ODB strain in a mixed genetic background due to epididymal and testicular dysfunction (see EMMA strain EM:12917). Breeding history | Loxl2 cDNA followed by a GFP reporter gene and an upstream PGK-neo cassette flanked by two loxP sites was introduced by homologous recombination in the endogenous Gt(ROSA)26Sor locus. ES clones positive for the mutation were microinjected/aggregated into receptive blastocysts/morulae and chimeras obtained. These animals were then crossed with cre recombinase-expressing mice to check for germline transmision and, at the same time, obtain animals constitutively expressing extra amounts of Loxl2. The resulting heterozygous animals were intercrossed and animals harbouring two Loxl2 overexpression alleles were obtained. Homozygous females in a mixed genetic background were intercrossed with C57BL/6J males for 12 generations to obtain animals harbouring two Loxl2 overexpression alleles in a pure C57BL/6J background. |
