Genetic information | Mice lack Mgll expression specifically in white and brown adipose tissue. MGL flox: Transfer of a targeting vector harbouring a loxP-flanked neomycin resistance gene cassette upstream of exon 3 and an additional loxP sequence within intron 4 of the murine Mgll gene (ENSMUST00000113585). The vector was introduced into HM-1 ES cells via electroporation. Clones that underwent homologous recombination were injected in C57BL/6 mouse blastocysts. Chimeric animals were bred with C57BL/6 mice. AdiQ-Cre: The bacterial artificial chromosome RP23 90G21 containing at least five mouse genes was modified by replacing the starting ATG and 222 bp of the adiponectin gene with the starting ATG and coding sequence of a cre recombinase gene. |
Phenotypic information | Homozygous:Homozygous MGL flox/AdiQ-Cre mice lack Mgll expression specifically in white and brown adipose tissue. This was proven by qPCR and immunohistochemistry. Mice are born within Mendelian frequency and both male and female mice are fertile. Compared to cre-negative littermates the mice do not show differences in body weight, lean or fat mass. No differences in glucose metabolism (fasted glucose and insulin, IpGTT, IpITT) were observed. However, male mice fed a high-fat diet for 24 weeks show increased plasma levels of TNFa, IL-10, and CXL1, but no differences for IL-6. Furthermore, the concentration of some acylcarnitines and glycerophospholipids was found reduced in white adipose tissue of mice fed a control or high-fat diet.Heterozygous:Heterozygous MGL flox/AdiQ-Cre mice are born within Mendelian frequency and both male and female mice are fertile. For these mice, no obvious phenotype was observed. Mice were not characterized any further. |
