Genetic informationThe murine Dusp9 locus was targeted by the introduction of loxP sites flanking exons 3 and 4 and the insertion of a positive selection marker (puromycin resistance) flanked by Frt sites downstream of exon 4 and immediately 5' to the loxP site within the 3' untranslated region. After electroporation into Art B6/3.5 (C57BL/6NTac) ES cells and puromycin selection, clones carrying the correctly targeted allele were identified and used to generate chimeric mice. Chimeric mice were then crossed with the C57BL/6-Tg(CAG-Flpe)2Arte Flp-deleter strain, resulting in recombination and removal of the puromycin selection marker to generate the conditional allele (Dusp9-fl).

Breeding historyBetween 2012 and 2014 DUSP9-fl mice have been backcrossed to C57BL/6J wild-type mice for 7 generations. Since 2014 there has been no backcrossing, just random matings from within the colony. No direct sibling matings are recorded, but the colony is very small (only 2 pairs or trios) so randomising the breeding to avoid pairing closely related animals would not have been possible. There have been 13 generations of these matings.
