Genetic information | Full-length human TNFalpha cDNA (Accession number NM_000594) was cloned behind the Tet-responsive P-tight promoter in the plasmid pTRE-Tight (BD Biosciences Clontech). The 3 prime UTR region of TNFalpha was replaced by the SV40 PolyA site, to avoid post-transcriptional regulation. The TNFalpha expression cassette under the Tet-inducible promoter was injected into the pronuclei of 0.5d FVB/N mouse oocytes. A positive Tg_ihTNFalpha founder was then crossed with transgenic mouse line hnRNPA2B1-rtTA2S-M2 (of C57BL/6 background) that was published earlier by Katsantoni et al. (2007). By further inbreeding, homozygous Tg_rtTA2/Tg_ihTNFalpha double transgenic mice were established and referred as ihTNFtg (see Retser et al.,2013 and Wixler et al., 2015). |
Phenotypic information | Homozygous:The mice are fertile and do not display any abnormalities without doxycycline stimulation. On doxycycline administration with drinking water the inducible ihTNFtg mice developed an inflammatory arthritis- and psoriasis-like phenotype with fore and hind paws and the skin at head and neck area being prominently affected. The formation of “sausage”-like digits with obligatory involvement of the distal interphalangeal joints and nail malformation is very characteristic. Synovial hyperplasia, enthesitis, cartilage and bone alterations, formation of pannus tissue as well as inflammation of the skin epidermis and nail matrix appeared as early as one week after the treatment of mice with doxycycline and became aggravated over time. The abrogation of hTNFalpha by the removal of doxycycline after few weeks of stimulation resulted in a fast resolution of the most accomplished macroscopic and histological disorders.Heterozygous:as homozygous |
