Service Description:

The Animal Modelling Facility (AMF) at the Netherlands Cancer Institute (NKI) provides all services related to the generation of genetically engineered mice (GEM) based on zygote manipulation by CRISPR/Cas9 procedures, including full-service covering the design and all hands-on steps. The AMF keeps track of recent technological developments and tests their usefulness for improving model generation. These include the application of CRISPR/Cas technologies in combination with Adeno Associated Viral vectors for the targeted introduction of larger inserts up to 4kbp.

Included in the Service:

After consultation with the researcher the AMF designs the genetic modification strategy including the setup of efficient genotyping protocols. Using the first generation of mice (founder) carrying the desired modification the AMF will set up breeding procedures to obtain the F1 generation to establish the genetically modified strain the for delivery to the researcher and long term cryopreservation. If needed the AMF will also enable revitalisation of the strains.

Additional Support:

The AMF provides the services of modulating ES-cells for in vitro studies and produce viruses for somatic modelling.

The Animal Modelling Facility (AMF) at the Netherlands Cancer Institute (NKI) consists of a fully equipped molecular biology facility for all the procedures involving assembly and validation by sequencing of DNA constructs involved in each zygote embryo manipulation. Also, genotyping of the mice derived from embryo manipulation and transfer is performed in this facility. In the Animal Facility the AMF has 1 room for embryo manipulation including 2 tissue culture incubators and 4 embryo manipulation stages. In addition, the AMF has 2 animal rooms in which mice are housed in individually ventilated cages at SOPF conditions.
