Phenotypic information | With this number of backcrosses (ca. 5) the floxed mice show some phenotype, since the floxed Il6 gene is expressed less than in normal wild-type mice upon endotoxin administration. We have already produced some conditional KO mice with several cre-expressing mouse lines and there is a clear difference between floxed and KO mice regarding Il6 levels. |
Breeding history | Neo cassette was removed by crossing with flp-expressing mice once one of the chimeras was found to have germ line transmission upon crossing with C57BL/6. The flp gene was removed upon crossing with C57BL/6. Finally, offspring mice positive for the floxed gene were crossed with each other and a homozygous line of these floxed mice is maintained that way in the general animal services of this university. We have started to further backcross this line with C57BL/6 in the Animal Physiology unit; in total we are about in the fifth generation. |
