Genetic informationA targeting vector containing a 4.6 kb XbaI/SmaI genomic fragment as the left homology arm, a 4 kb SmaI/HindIII genomic fragment as the right homology arm, a loxP-flanked neo selection cassette (L2neo) introduced into a SmaI site in intron 5 of the p55TNFR (Tnfrsf1a) gene between the two homology arms and a PGK-tk negative selection marker at the 3' of the targeting vector, was transfected into ES cells for the generation of the p55cneo allele.
Phenotypic informationMutant mice carry a conditional gain-of-function allele (p55TnfRcneo) obtained by introducing a loxP-flanked neomycin resistance cassette in intron 5 of the murine p55TnfR (Tnfrsf1a) gene. p55TNFR expression is inhibited by the presence of the neomycin cassette but it is restored upon Cre-mediated neo excision.
