Genetic informationA pGK1neo cassette replaces the alpha-helical coiled coil sequence of exon 4 (essential for the formation of functional trimeric receptors) of the endogenous Msr1 gene. The construct was electroporated into A3-1 embryonic stem (ES) cells of 129X1 origin. Correctly targeted ES cells were injected into C57BL/6J blastocysts and transplanted into the uteri of ICR females. The resulting chimeric males were backcrossed to ICR females and heterozygous mice were bred to generate homozygous mice. Subsequently mutant mice were backcrossed to C57BL/6J for 12 generations. Around 1998, Dr. Hiroshi Suzuki and Dr. Tatsuhiko Kodama sent these mutant mice to Prof. Siamon Gordon, Univ. of Oxford.
Phenotypic informationHomozygous:
Homozygotes are viable and fertile. Liver sections treated with antibodies against type I and type II macrophage scavenger class A receptors show no protein staining. Homozygous mice have less severe lesion development in Apoe-deficient atherosclerosis model. Peritoneal macrophages have a reduced mLDL uptake in vitro. Homozygous mice have increased susceptibility to various pathogens. These mice may be useful in studies of immunity and host defence, cholesterol transport in macrophages, macrophage-derived foam cells, and atherosclerosis.

Same as wild-type.
Breeding historyMsr1 knock-out mice were backcrossed to a C57BL/6J line for 10 generations and the established homozygous line has been maintained as brother x sister mating within Univ. of Oxford facility. They breed just as good as the background strain with average litter sizes of 5/7 pups and the health status has remained consistent for many years.
